Even though it's a flash game, I always liked Gemcraft series. It has some very interesting concepts, the main one being that monsters and gems you put into towers have different hues and based on the difference between them, they deal either increased or reduced damage. So you have to mix gems correctly to have corresponding hues to the incoming enemies to deal maximum damage.

It also has quite an expansive skill tree in each installment, allowing for a variety of play styles.

I'd recommend Kingdom Rush

It's quite fast pace and requires you to use active abilities. So if you're looking for a game to just lean back and watch this is not for you.

Kingdom Rush and Defender Quest are classics. Kingdom Rush is really the pure paradigm of tower defense, Defender Quest adds a bit of humour and personalized characters. These two games should suffice for (good) pure core tower defense gaming.

The Anomaly series is sweet, but it's a reverse tower defense (tower offense?), until the last opus, which is standard defense, but I haven't played it. These games are visually gorgeous.

And you have fun tower defense minigames in the incredible adventures of van helsing. They're just optional breaks from the diablo-like gameply, but, heck, I like the incredible adventures of van helsing, so, I have to mention this.

I second Kingdom Rush, it was fantastic. Also second the Orcs Must Die series. I also played Sanctum. It's serviceable. It's a bit meh.
